Although both the answer and definition are singular nouns, I don't understand how one could define the other.

'with capek's play for first time in mythical place' is the wordplay.
'with' says to put letters next to each other.
'capek's play' becomes 'rur' (I've seen this in another clue).
'for' is a charade indicator (letters next to each other).
'first' becomes 'i' (Roman numeral for one or first).
'time in mythical place' becomes 'tania' (I am not sure about this - if you are sure you should believe this answer much more).
'rur'+'i'='ruri'
'ruri' next to 'tania' is 'RURITANIA'.
