Although both the answer and definition are singular nouns, I cannot understand how one could define the other.

'is not entirely all there after demolition ultimately' is the wordplay.
'not entirely' means to remove the last letter (most but not all of the word).
'all there' becomes 'sane' ('not all there' can mean insane).
'after' says to put letters next to each other.
'ultimately' says to take the final letters.
The final letter of 'demolition' is 'n'.
'sane' with its final letter taken away is 'san'.
'is'+'san'='issan'
'issan' after 'n' is 'NISSAN'.
