Although both the answer and definition are singular nouns, I cannot see how they can define each other.

'leading lady in' is the wordplay.
'leading' becomes 'ur' (I can't explain this - if you can you should give a lot more credence to this answer).
'lady' becomes 'she' (I've seen this before).
'in' is an insertion indicator.
'ur' going around 'she' is 'USHER'.
